Abstract

U ovom preglednom radu cilj je bio prikazati temeljne biokemijske karakteristike bioaktivnih lipida, isprepletenost njihovih složenih metaboličkih puteva, kao i njihove konačne učinke. Bioaktivni lipidi nastaju kao derivati arahidonske kiseline, ili cijepanjem drugih elemenata stanične membrane. Dugo vremena su lizofosfolipidi i sfingolipidi smatrani samo strukturnom komponentom stanične membrane, međutim današnje spoznaje otkrivaju njihov izravan utjecaj na signalne putove stanica urođene i stečene imunosti. Eikozanoidi, koji nastaju kao derivati dokozaheksaenonske ili eikozapentaenonske kiseline, svojevrstan su sinonim za bioaktivne
lipide, te svojim proupalnim i protuupalnim učincima imaju važno mjesto u upalnom odgovoru organizma. U emocionalnoj regulaciji, kao predstavnici bioaktivnih lipida, najvažniji i najistraženiji su endokanabinoidi. Navedene spoznaje imaju velik potencijalni značaj jer čine bioaktivne lipide privlačnom metom za liječenje neuroinflamatornih i neuroimunoloških poremećaja, uključujući Alzheimerovu bolest, multiplu sklerozu, amiotrofičnu lateralnu sklerozu, traumatsku ozljedu mozga, anksioznost i depresiju.

Abstract (english)

The aim of this thesis was to show the basic biochemical characteristics of bioactive lipids, the intertwining of their complex metabolic pathways, and their target effects. Bioactive lipids are formed as arachidonic acid derivatives or by cleavage of other cell membrane elements. For a long time, lysophospholipids and sphingolipids were considered only a structural component
of the cell membrane, however, recent findings reveal their direct influence on signaling pathways within cells of innate and adaptive immunity. Eicosanoids, which are formed as derivatives of docosahexaenoic or eicosapentaenoic acid, are a kind of synonym for bioactive lipids, and their pro-inflammatory and anti-inflammatory effects have an important place in the
inflammatory response of the organism. The most important and most researched investigated bioactive lipids involved in emotional regulation are endocannabinoids. These findings are of great potential significance because they make bioactive lipids an attractive target for the treatment of neuroinflammatory and neuroimmunological disorders, including Alzheimer’s
disease, multiple sclerosis, amyotrophic lateral sclerosis, traumatic brain injury, anxiety and depression.
