Abstract Vrat maternice važan je čimbenik plodnosti i reproduktivnog zdravlja žene. Nabori u
sluznici cervikalnog kanala, tzv. endocervikalne kripte ili endocervikalne žlijezde,
proizvode cervikalnu sluz. Po svom biokemijskom sastavu cervikalna sluz je
glikoproteinski gel. Njegova proizvodnja hormonski je regulirana, o čemu ovise
njegova biokemijska i biofizička svojstva. Cervikalna sluz osim zaštitne funkcije ima
važnu ulogu i u plodnosti jer prihvaća, hrani i čuva spermije te im pomaže u prijenosu
do jajne stanice kako bi došlo do oplodnje. Mehanizam stanične regulacije
proizvodnje sluzi različitih svojstava, te njihova otpuštanja u lumen vrata maternice
tek je djelomično poznat.
Cilj ovog istraživanja bio je razjasniti dio mehanizma sekrecije cervikalne sluzi
vodeći se predloženim modelom sudjelovanja neurotransmitera GABA u hormonskoj
regulaciji izlučivanja sluzi. Istraživanje je provedeno na fiksiranim uzorcima tkiva
vrata maternice dobivenih nakon histerektomije (operacijskog zahvata uklanjanja
maternice).
Najvažniji rezultati ovog istraživanja pokazuju prisutnost molekula GABA-ergičkog
signalnog puta u žljezdanom epitelu tkiva vrata maternice. Rezultatima je utvrđeno da
prisutnost receptora A za neurotransmiter GABA ne korelira s fazom ciklusa u
kojemu je bila pacijentica u trenutku uzorkovanja. Analizom rezultata istraživanja
pokazalo se također da je prisutnost receptora A za neurotransmiter GABA češći u
gornjem dijelu endocerviksa nego u donjem. Ovi rezultati svakako pridonose raspravi
o ulozi signalnog puta GABA-e u izlučivanju sluzi. Fiksirani uzorci tkiva vrata
maternice dobiveni nakon histerektomije pristupačan su i dobar model za
preliminarno ispitivanje hipotetskih prijedloga o mehanizmu regulacije stvaranja i
izlučivanja endocervikalne sluzi, ali ne i za donošenje definitivnog zaključka o
predloženim hipotezama.
Abstract (english) The cervix is an important factor in a woman's fertility and reproductive health. The
folds in the mucosa of the cervical canal, the so-called endocervical crypts or
endocervical glands, produce cervical mucus. By its biochemical composition,
cervical mucus is a glycoprotein gel. Its production is hormonally regulated, which
depends on its biochemical and biophysical characteristics. Cervical mucus, in
addition to its protective function, plays an important role in fertility as it accepts,
nourishes and protects sperm and helps transfer it to the ovum for fertilization. The
mechanism of cellular regulation of mucus production of different properties, and
their release into the lumen of the cervix, is only partially known.
The aim of this study was to elucidate part of the mechanism of cervical mucus
secretion, guided by the proposed model of the involvement of the neurotransmitter
GABA in the hormonal regulation of mucus secretion. The study was performed on
fixed samples of cervical tissue obtained after hysterectomy, (surgical removal of the
uterus).
The most important results of this study show the presence of a GABA-ergic
signaling pathway molecules in the glandular epithelium of cervical tissue. The
results established that the presence of receptor A for the neurotransmitter GABA did
not correlate with the phase of the patient's cycle at the time of sampling. Analysis of
the results of the study also showed that the presence of receptor A for the
neurotransmitter GABA is more common in the upper endocervix than in the lower
one. These results certainly contribute to the debate about the role of the GABA
signaling pathway in mucus. secretion. Fixed cervical tissue samples obtained after
hysterectomy are an affordable and good model for preliminary examination of
hypothetical proposals for the mechanism of regulation of endocervical mucus
formation and secretion, but not for reaching a definitive conclusion on the proposed
hypotheses.
