Abstract

Žljezdani epitel ljudskog endocerviksa izlučuje sluz koja se tijekom ženskog mjesečnog spolnog ciklusa mijenja te u vrijeme ovulacije poprima svojstva koja omogućuju spermijima preživljavanje i oplodnju. Mehanizam hormonske regulacije tih promjena još je nepoznat. Na temelju naših prijašnjih rezultata koji su pokazali prisutnost GABA-ergičkih molekula GABAAR i GAD u žljezdanim stanicama endocerviksa te sličnosti sa žljezdanim epitelom dišnog sustava predložena je uloga GABA-ergičkog sustava u hormonskoj regulaciji izlučivanja i oblikovanja sluzi. Cilj istraživanja, provedenog na osamnaest postoperativnih uzoraka ljudskog endocerviksa, bio je ispitati povezanost intenziteta izražaja GABA-ergičkih molekula i CFTR-a s koncentracijom estradiola u krvi, a izražaj GABA-ergičkih molekula i s intenzitetom izlučivanja mucina. Rezultati istraživanja pokazuju da izražaji GABA-ergičkih molekula nisu povezani s razinom estradiola u krvi, a nije pokazana ni povezanost s intenzitetom izlučivanja sluzi. Ispitivanje povezanosti izražaja CFTR-a s koncentracijom estradiola pokazalo je porast izražaja CFTR-a u apikalnom dijelu epitela uz porast estradiola, kao i da porast progesterona poništava ovaj učinak. Ovi rezultati potvrdili su pretpostavku da CFTR može imati ulogu u hormonskoj regulaciji formiranja sluzi, te da važnu ulogu ima njegova lokalizacija u stanici. Pozitivna korelacija CFTR-a i VAMP8 ukazuje na moguću ulogu VAMP8 u regulaciji lokalizacije, a time i djelovanja CFTR u endocervikalnom epitelu.

Abstract (english)

The secretory epithelium of the human endocervix secretes mucus that changes during the menstrual cycle and, at the time of ovulation, gains properties that allow sperm survival and fertilization. The mechanism modifying the hormonal regulation of these changes is still unknown. Our previous research has shown the expression of GABA-ergic molecules GABAAR and GAD in glandular endocervical cells. Based on the similarity to the respiratory system glandular epithelium, we suggested a role of the GABA-ergic system and CFTR in the hormone regulation of mucus secretion and formation. The aim of the study, performed on eighteen postoperatively obtained samples of human endocervical tissue was to investigate the correlation of CFTR and GABA—ergic molecules expression with blood estradiol concentration, and the correlation of GABAAR and GAD with the intensity of mucin secretion. The results of the study show that the expressions of GAD and GABAAR are neither related to the serum estradiol level, nor to the intensity of mucus secretion. Positive correlation of estradiol and CFTR expression in the apical part of the epithelium, which is attenuated by rising progesterone, confirmed the hypothesis that CFTR may play a role in hormonal regulation of mucus formation.
